Hampshire seamer Dominic Cork will make his first return to Old Trafford since his release by Lancashire last summer.
Lancashire will be without injured opener Tom Smith, while Andrew Flintoff and James Anderson are away on England preparing for Ashes duty.
But Sajid Mahmood should be back from England Lions duty and Stephen Parry has a case for replacing Gary Keedy in the light of his recent Twenty20 form.
Mal Loye also looks set for a first one-day start of the summer.
After missing the first two months of the season, he then failed to get a bat in Lancashire's last two Twenty20 games.
Lancashire will be wary of Pakistan leg-spinner Imran Tahir after their two previous encounters with him in Championship cricket.
He took 6-109 to bowl out Lancashire and set up his side's 10-wicket win at Liverpool last month. And he did even better at Old Trafford last summer, finishing with match figures of 12-189.
Hampshire look set to play the same XI that beat Essex in last weekend's final Twenty20Cup group game, which would mean Hamza Riazzudin and Billy Taylor missing out.
The winners will meet either Moores' old county Sussex or Gloucestershire in the final at Lord's on 25 July.
